WI: Pedro de Mendoza dies in 1528?

Pedro de Mendoza was one of the most important Spanish conquistadores in South American history. In 1534 he embarked to the continent and was responsible for the short-lived first founding of Buenos Aires, and whose failed expedition would inadvertently lead to the founding of Asunción by the survivors of Buenos Aires's destruction, the capital of modern Paraguay. Due to the importance of Asunción and its role in making much of modern-day South America, it begs the question of how would South America develop if Pedro de Mendoza died before he could offer his services to the Spanish crown.
But how he would die? Shyphillis. He contracted it in the Italian War of 1527 after fighting for the Habsburgs, and was severely ill with it. Assuming that he dies of syphillis in 1528, how does South America develop?
