The Challenge is easily spelled out. The challenge already has a problem which is a combination of regional goal for the only kingdoms/empires with any sort of greek focus being in the eastern Medi. So I guess the challenge needs three things:
the POD should be in the 1100's as that should allow the rest of the nations which existed in western Europe to hopefully evolve relatively as normal at least in to the 13th-14th century.
That means Las Navas De Tolosa will occur regardless and with a Christian victory.
There will be no Angevin empire either, Richard the Lion Heart will also die due to a mangled surgery to remove a bolt from his shoulder.

And most importantly, a bumbling fool given the funds to explore a faster route to India will end up discovering the island in the Caribbean.
These are things that Must occur, now that those standards have been laid down. How exactly could an a nation based in the balkans/ anatolia end up sending any men or resources as far west as the Americas(Or whatever they are called in this ATL)

Does a Venetian colony with a Greek majority population count? A 12th century POD would probably change too many things.

The problem is that generally only Atlantic and Baltic countries cared about the Americas because they were jealous of the Mediterranean countries' trade advantage. Nobody in the Balkans or Anatolia needed an alternate route to India and China because they already had the main route. Venetian and other Italians are heavily involved with those Atlantic Empires, but only under the employ of foreign states.

Maybe a more successful Catalan Company results in the Aragonese getting a permanent hold in Greece. Aragon unites with Castile as in OTL except Castile doesn't get the monopoly on transatlantic imperialism. Greeks are recruited on mass into the Spanish navy and there are Greek majority towns established throughout the world?

Alternatively, Stefan Uroš IV Dušan, Tsar of the Serbs and the Greeks has a less incompetent successor and the Ottomans are too ravaged by Timur to stop the creation of a stable Serbo-Greek Empire and... No, they still wouldn't bother leaving the Mediterranean without some kind of unmaintainable Venetian and Turkish embargo.

Does it have to be the colony of a Greek state, or just a large Greek population, because the latter is a whole hell of alot easier than the former.
